The utility of ultra-deep RNA sequencing in Mendelian disorder diagnostics

RNA sequencing has emerged as a powerful tool for Mendelian disorder diagnostics, but the optimized sequencing depth has not been determined. Using Ultrahigh- depth RNA sequencing (up to 1 billion reads), we improved detection of low-abundance genes and transcripts, created resources to guide sequencing depth, and revealed new insights for genetic diagnosis.
